Isaiah was born in Saybrook, Middlesex, Connecticut on 29 December 1788 to Joshua Post, age 45, and Anna Jones, age 46.

In October 1811, at the age of 22, Isaiah married Lydia Doane and had four children, including Lydia Harriet Post. Lydia, his wife, died in 1835 when Isaiah was 46. He remarried six years later at the age of 52.

Isaiah died on 9 April 1873, in Westbrook, Connecticut, when he was 84 years old. He was buried in Westbrook.
